The drug Tianeptine is a research chemical classified as an atypical antidepressant which is an antidepressant that acts in an atypical manner compared to the majority of other antidepressant medication. Other atypical antidepressants include nefazodone, trazodone, mirtazapine, bupropion and agomelatine. It also has anxiolytic, or antianxiety, effects. Uniquely, the drug relatively lacks the sedative, cardiovascular and anticholinergic side effects that similar substances have shown.
Studies show that tianeptine also acts as a μ-opioid receptor (MOR) atypical agonist. μ-opioid receptors (MOR), also known as μ-opioid peptide (MOP) receptors, have a high affinity for beta-endorphin and enkephalins, but a very low affinity for dynorphins. Morphine, the main psychoactive alkaloid in opium, is a prototypical μ-opioid receptor agonist. Studies showed μ-opioid receptors induce euphoria which caused concerns for abuse.

In the 1960’s, the French Society of Medical Research discovered and then patented the drug. Asia as well as several countries in Europe currently prescribe Tianeptine. Laboratories Servier SA manufactures the drug under the name of Coaxil. It is available for research purposes only in the United States.
Tianeptine has has several clinical trials over the years to see the effects on a wide variety of ailments such as depression, anxiety, irritable bowel syndrome, asthma, pain due to fibromyalgia and even attention deficit hyperactivity disorder (ADHD). Past the potential applications that these clinical trials showed, clinical trials also showed several dangers and adverse reactions. The most notable of these was the dangerous interaction when monoamine oxidase inhibitors (MAOIs) are currently being taken or have been taken within at least 14 days prior. This can cause hyperthermia (high body temperature), convulsions, cardiovascular effects (including cardiovascular collapse and hypertension) and even death.
